Have you ran postmap after upgrading? Perhaps the binary format has changed. 

Try running postmap on the file directly and add one or more -v switches.

That was true for 2.3 as well. Domains that lack real accounts shouldn't 
be listed on the mydestination line. Postfix will automatically 
recognize that it is the designated recipient for domains listed in the 
virtual tables.

No, virtual aliases are appropriate for what you're doing. canonical is 
for rewriting headers.
